Advancements in MESF Beads for Enhanced Fluorescence Quantification in Flow Cytometry The document discusses advancements in the development of MESF (Molecules of Equivalent Soluble Fluorochrome) beads for improved fluorescence quantification in flow cytometry. It highlights the importance of MESF beads in standardizing fluorescence measurements across instruments and experiments. The research introduces new nano-sized MESF beads incorporating various fluorescent dyes such as Pacific Blue®, SuperNova, FITC, PE, and APC. These beads are designed to have low background fluorescence and known dye content, making them suitable for small particle flow cytometry. Preliminary testing using the CytoFLEX nano Flow Cytometer demonstrated their potential for accurate quantification of dye molecules per bead. The study establishes a reliable methodology for MESF value calculation and shows how these beads can be used to determine fluorescence detection limits in biological samples. Although not yet commercially available, the new MESF beads are expected to become essential tools for instrument calibration and assay standardization in advanced flow cytometry applications.
